- 【I】specimen pretreatment
- ① We collect the food (sample) to be tested. The instruments used for sampling (tweezers, scissors) should be sterilized or sterilized by alcohol or flame sterilization.
- ② Set the weighing processing sample collection bag stand on the balance. Insert the Stomac bag (E-Mix Homogenize bag) into the balance bag stand and open it. Tare the balance and weigh 10 g of the sample into a Stomach bag (instruments must be sterilized at this time). Then add sterile diluted water OR-90 (90 mL) to make a 10 fold diluted sample solution.
- ③ Mix a Stomac bag containing 10 g of the sample + sterile diluted water OR-90 (90 mL) with a Stomacker (bag mixer E-Mix).
- 【II】serial dilution
- ④ Dilute further using 10 times dilution. Place the treated Stomac bag on the food sample stand as shown in the photo and use a micropipette to aspirate 1 mL.
- ⑤ Inject 1 mL of the aspirated 10 × diluted sample solution into sterile diluted water OR-9 (9 mL) to complete the 100 × diluted sample solution (Repeat as shown in the figure if necessary to make 1000 or 10,000 times diluted sample solution.).
- [III] Sample inoculation
- ⑥ Drop 1 mL of the 10, 100 and 1000 times diluted sample solution onto 2 sheets of Petri FilmTM, and cover with the film.
- ⑦ Press lightly with a special tool (spreader) and mold into a circle.
- ⑧ Once it has solidified into a gel, put it in an incubator and incubate at the specified temperature and time. *Petri FilmTM for coliform bacteria and E. coli is difficult to determine after incubation if air enters the film. Please be careful.
- ⑨ After incubation, measure with a colony counter pen. 30~300 colonies is an appropriate number. The average value is obtained for each dilution and the number of bacteria is calculated by multiplying the dilution ratio.
- ⑩ Put the used Petri FilmTM in a sterilization bag, sterilize by high pressure steam, and discard.
- Bacterial count calculation method [Example]
- ① Average number of colonies (155+180) ÷2=167.5≒168 in 2 sheets of 10 × dilution solution
- ② Mean number of colonies multiplied by dilution factor: 168 x 10=1680
- ③ Multiplier: 1680=1.68 x 10^3
- Bacterial count: 1.7×10^3 CFU/mL
- When the dilution ratio is 10: 1 sheet/155, 2 sheets/180
- When the dilution ratio is 100: 1 sheet/20, 2 sheets/17
